Specific Description  Members of the glial cell line-derived neurotrophic factor (GDNF) family, including GDNF and neurturin (NTN), play key roles in the control of vertebrate neuron survival and differentiation. Physiological responses to NTN require the presence of a novel glycosylphosphadidylinositol linked protein NTNRalpha, which is a cell surface receptor for NTN. The cDNAs encoding NTNRa from human, rat, chicken, and mouse have been cloned recently. NTNRalpha was also termed GDNFRbeta, Ret ligand 2 (RETL2) or TGFbeta-related neurotrophic factor receptor 2 (TrnR2) and nominated as GFRalpha-2 recently. GFRalpha-2 binds NTN and mediates activation of RET receptor tyrosine kinase by both NTN and GDNF. Thus, NTN, GFRalpha-2, and the Ret PTK form a complex to transduce NTN signal and to mediate NTN function.

A new member of the GDNF family was recently identified and designated persephin. Physiological responses to these neurotrophic factors require two receptor subunits, the novel glycosylphosphadidylinositol linked protein GFRalpha and Ret receptor tyrosine kinase GFRbeta (2). Following the findings of GFRΑ-1 and -2, a novel receptor in GFRalpha family was identified very recently from human and mouse and designated GFRalpha-3. GFRalpha-3 binds persephin, thus, persephin, GFRalpha-3, and Ret PTK form a complex to transduce persephin signal and to mediate persephin function.

Specific Description  The tumor necrosis factor (TNF) and TNF receptor (TNFR) gene superfamilies regulate numerous biological functions including cell proliferation, differentiation, and survival through regulating the activation of the transcription factor NF-kappaB and various mitogen-activated protein kinases (reviewed in 1). Nerve growth factor receptor (NGFR) was one of the earliest characterized members of this family (2). Also known as the low-affinity receptor p75NTR, this receptor is involved in several diverse functions such as apoptosis, neurite outgrowth during development, and myelination (reviewed in 3). Its ligands include NGF, brain-derived neurotrophic factor (BDNF), NT3, and NT4 (4). NGFR can also associate with other NGF receptors such as Trk through the cytosolic and transmembrane domains and thus can function as a co-receptor that refines Trk affinity and specificity for neurotrophins (5). Finally, upon binding of various neurotrophins, NGFR associates with tumor necrosis factor receptor-6 (TRAF6), suggesting that it can potentially function as a signal transducer for NGF signals through NGFR (6).
